DEVELOPMENT OF MUSSEL-SPECIFIC EXPRESSION VECTORS SUITABLE FOR TRANSGENIC ORGANISMS : IMPLICATION IN THE ESTABLISHMENT OF MOLLUSC CONTINUOUS CELL LINE

Objective

The main goal of this project is to develop basic means for in-depth understanding of viral diseases affecting commercially exploited molluscs. This will be achieved through the development of functional mollusc specific expression vectors and subsequently by the obtention of mollusc continuous cell lines. We will study Mytilus edulis since it is widely used as an experimental model. The project will comprise six main experiments; some of them can be initiated independently and therefore allow some flexibility in the realisation of this project. The goal is to be reached through a collaborative effort between the participants using complementary approaches.

Experiment 1: Cloning of mussei genomic DNA. Isolation and characterization of mussel genes.
Experiment 2: Mussel specific expression vectors.
Experiment 3: Establishement of primary cell culture system from the mussel Mytilus edulis.
Experiment 4: Expression of oncogenes in mussel cells.
Experiment 5: Isolation of genetic markers.
Experiment 6: Stable cell lines from Mytilus edulis.
