Interaction of calcium and a-latrotoxin in the stimulation of neurosecretion

Obiettivo

Release of neurotransmitters from neuronal cells is a basis for neuron-neuron communication and, in general, for the working of the brain. The research will use the neurosecretogogue a-latrotoxin (a-LTX) and in vitro culture of chromaffin cells of neuronal origin to study neurosecretion. a-LTX induces secretion of catecholamine in calcium-dependent manner from both intact and permeabilised cells. The toxin interacts directly with the neurosecretory machinery.

The research unites expertise in toxin handling and protein biochemistry at the Shemyakin Institute with expertise in state-of-the-art calcium measurement and imaging techniques at University College London (UCL). The participation of scientists from both research groups will give them access to resources not yet available at the Shemyakin Institute and will facilitate the setting up of such resources at a future date.

The aim of the research is to identify the molecular basis of the interaction between calcium and a-latrotoxin using a combination of cell physiology, cell biology and biochemistry. This will lead to a fundamental insight into the process of neurotransmitter release, and thus to an understanding of brain mechanisms.
