Structure and function of the chromosomal activator protein HMG1

An understanding of cellular processes at the molecular level is the basic knowledge that underpins diagnosis and therapy in human medicine. Gene transcription and its control is a key process, not least because cancer results from aberrant gene expression. The protein HMG1 plays a general, but as yet poorly understood, role in gene transcription and this research project will further the understanding of its function. Since it bends through a large angle on binding, it exhibits very special properties. HMG1 contains two HMG boxes, a new DNA binding motif found in other transcriptional activators, and therefore all the findings should have a broader application.

Mapping the location of HMG1/2 in the genome will be a major task. Therefore monoclonal and polyclonal HMG 1 antibodies will be produced and checked using the Western blot method. DNA / HMG1 complexes will be transfected into mammalian cells and the efficiency of the transfection will be studied. The structure of the DNA / HMG1 complex will be investigated usibng optical (CD) and biochemical (EMSA) methods. The complex will also be studied by measurement of the fluorescent intensity and of the fluorescent resonant energy transfer.
