Recombinant polypeptides for antigenic mimicry of bacterial surface polysaccharides

Objective

We have recently demonstrated that anti-idiotypic antibodies and peptides can mimic efficiently polysaccharide antigens and that protection through vaccination is possible. To extend this approach in order to obtain reagents with higher affinity and industrial utility we propose the following project. The main goal is the production of polypeptide structures which mimic polysaccharide epitome exploiting qualified knowledge in all known techniques for producing peptide- and antibody-based surrogate antigens. The three pathogens selected are appropriate for this aplication for distinct reasons:
1) Brucella vaccination with whole S-LPS makes epidemiological control by serology impossible,
2) Vibrio S-LPS exhibit poor immunogenicity and high crossreactivity,
3) for S.pneumonie conjugate vaccines will not provide a complete solution for the problems with the current vaccine.
