Effects of pahs in environmental pollution on exogenous and endogenous dna damage

This project will evaluate the hypothesis that Paths are the major source ofgenotoxic activities of organic mixtures associated with air pollution. Such exposures result in exogenous DNA damage but the importance of endogenous damages unknown. Exposure of 4 populations exposed to Paths will be measured and lifestyle factors determined. Exogenous exposure will be measured by determination of adducts, protein induction, and gene expression, and endogenous damage by measurement of oxidative DNA modification. Effect will be monitored by determination of chromosome aberration, and susceptibility by genotyping a wide variety of enzymes involved in carcinogen metabolism. Model in vitro cell systems will be proposed for studying the impact of Paths in complex mixtures of different air pollution origin. The project will define which biomarkers should be used for risk assessment of environmental exposure to Paths.
