Pathophysiology and prevention of lactococcus garvieae and streptococcus iniae infections in rainbow trout

Obiettivo

L. gravitate and S. inane infections represent the major cause of rainbow trout losses in several Mediterranean countries. Recently. This Gram-positive cockchafer also been associated with human infections. In trout, average mortality rates head to 50% during the hot season, due to the fact that conventional treatment are little effective. The massive use of antibiotic for controlling these diseases, besides being inadequate, is expensive and environmental not friendly. The development of preventive tools is therefore essential. The aim of the project into define valid methods to prevent the onset of these infections, through the generation of path physiological data that will be helpful for exploring the immune response in vaccinated fish. The availability of efficient preventive tools would allow to control L. gravitate and S. inane infections, with an improvement of aquaculture production in the European Union.
What generated provided new insights of the immunological and immunopathological machineries of fish versus infections sustained by pathogenic Gram-positive cocci. In additions, it provided novel scientific data regarding the virulence mechanisms sustained by these pathogens.

As a whole, the results may be summarized as follows:
WP1. Construction of host/pathogen interaction models In the present work, animal models were replaced with cell-lines models. In-vitro assays were successfully developed for studying the following issues: adhesion to cells, penetration into cells, intracellular survival, trafficking inside cells, cell damages inflicted by the pathogens.
WP2. Modulation of the immune response in diseased fish Basic science data regarding the immune system of fish and how it is modulated by pathogens were obtained. The major achievements were the following: - elucidation of the specific cell immune response, with major emphasis on PN-invasion and opsonin-dependent phagocytosis; - generation of data concerning the intestinal immune system, including the isolation and purification through a mechanical method of the trout leukocytes populations. WP3. Eliciting of a specific immune response. This part of the work was oriented to explore potential remedies to losses caused by Lactococcosis and Streptococcosis, with emphasis on the feasibility of specific vaccination strategies. The major achievements were the following: - demonstration that injectable vaccines assist in limiting losses; generation of laboratory parameters that allow correlation between specific immunity and resistance; highlighting the problematic of dip/oral vaccination.
