Objectif Aspergillus niger is the main microorganism used for enzyme production within DSM Food Specialties. The research proposal aims at the utilization of modern genomics technologies in order to rationalize strain and process improvement programs for enzyme production using A. niger as a host. Since DSM has sequenced the complete genome of A. niger, DNA micro arrays and proteomics will be used for studying the expression profile of all genes for various strains under various controlled fermentation conditions. Focus will be on identification of genes involved in heterologous gene expression and genes influencing fungal morphology. Knowledge on the (differential) expression of these genes will be used to construct new A. strains by over expression / deletion of specific genes or by Classical Strain Improvement (CSI) followed by targeted screening. Alternatively fermentation conditions will be adapted based on knowledge obtained after expression profiling. These experiments should give us information on bottlenecks in (heterologous) gene expression and genes involved in viscosity /morphology with the final goal to increase the yields in A niger fermentations. Strains expressing homologous genes and strains expressing heterologous genes will be compared. Low and high copy number strains will be examined in order to identify limiting factors at high copy numbers. Mutants obtained after CSI will be compared with parent strains in order to find new leads for strain and process improvement.
