Ras participation in neurological disease

Obiettivo

Participation of simultaneously nitrosylated and farnesylated Ras-protein in neuronal apoptosis during brain ischaemia and experimental allergic encephalomyelitis.
Ras proteins are proto-oncogene products that are critical components of signalling pathways leading from cell-surface receptors to the control of cellular proliferation, differentiation or cell death. Activation of Ras is associated with ischaemia/reperfusion damage of brain or with neuroinflammatory diseases. Post-translational modification is responsible for the proper functioning of Ras. The main scientific objective of the project is the investigation of role of nitrosylation on the membrane-binding properties of Ras and downstream effectors specificity of simultaneously (farnesylated and nitrosylated) modified isoforms of Ras.

The project is directed towards the following points:
(a) Purification, isolation, preparation, and investigation of biological activities of double modified isoforms (simultaneously nitrosylated and farnesylated) of Ras;
(b) Investigation of adhesive and phospholipid-binding properties of various modified isoforms of Ras using a highly sensitive integrated optical technique allowed detailed and sophisticated analysis of the entire binding phenomenology;
(c) Investigation of the action of farnesyltransferase and nitric oxide synthase inhibitors on the induction/prevention of neuronal damage during brain ischaemia and experimental allergic encephalomyelitis in rats.

The anticipated results will reveal the functionally active form of Ras which participates in different downstream processes of the cell. After successful completion of the project, it will be possible to recommend types of pharmacological drugs, which will diminish the apoptosis/necrosis of neuronal cells after ischaemia/reperfusion damage of brain or after neuroinflammation.
