Objetivo The project deals with a new class of MADS box proteins that were identified in Arabidopsis using the data of the genome sequencing project. The phylogenetic analysis shows that these proteins belong to the SRF-like type of the eucharistic MADS box regulatory factors, which are characterised by different conserved a residues in the MADS box and different structural features in the C-terminal part of the protein when compared to known plant MADS box proteins. The project aims to functionally analyse these new type of MADS box genes, concretely the representatives of the ALG29 and AGL30 clad. Arabidopsis plants will be transformed with over expression, RNA interference (for repression of a transcript) and VP16-GR fusion constructs. Transform ants showing interesting phenotype will be further molecularly and morphologically characterised. An Arabidopsis transposing population, available to the group of Dr. L. Colombo, will be screened for insert ional mutants in the genes of interest. For the selected candidates an experiment will be performed, using the VP16-GR Arabidopsis transform ants and the micro array, to identify target genes. The yeast two-hybrid system will be applied to identify a part of the protein responsible for dimidiation (by a deletion study), and the three-hybrid system to analyse a possible ternary complex formation amongst the new MADS box proteins. If one of these genes shows to have an interesting agronomical trait, it will be used to transform rice.
