Objetivo THE APPLICANT will get experience in the initiation and development of genetically transformed "hairy" roots, as well in the manipulation of cell cultures initiated from woody plants of dicotyledonous (Vitas) and coniferous origin (Taxes). Confirmation of transformation could be carried out by DNA analyses. He will also get instruction on and experience with the isolation and identification (chemical and chromatographic) of plant secondary metabolites, including HPLC protocols for quantitative analyses. THE HOST INSTITUTION will gain from the expertise and experience, which the applicant has in the areas of somatic embryogenesis, protoplast isolation/culture and calcium dynamics of plant cell cultures. This is of special interest in the case of Taxes baccarat where micro propagation and embryogenesis have proved difficult. Also protoplast isolation from metabolite-rich and fast-growing strains of Taxes could be extended to include protoplast fusion to develop hybrids of commercial interest.
