Workshop on "Systems biology of DNA-damage-induced stress responses"

The objective of this project is to co-finance a collaborative Workshop on "Systems biology of DNA damage induced stress responses" between researchers of the EU and the USA. The purpose of this Workshop is to develop better collaboration between researche rs in the EU and the US in a systems biology analysis of the principal pathways in the area of DNA-damage response. Heretofore, researchers focused on specific cellular compartments with regard to DNA-damage responses, such as cell cycle regulation, indu ction of apoptosis or DNA repair. However, work during the past few years has shown that there are strong interplays between specific steps in these major defence pathways and in order to fully understand cellular responses to DNA-damage, the cell must b e viewed as a large network of protein-protein and protein-DNA interactions that respond in a systematic way to DNA-damage. Researchers of the various DNA-damage response pathways have therefore recently embraced approaches in which global measurements of gene expression and the proteome can be combined with genome-wide screening of sensitivity mutants to develop an integrated view of how cells respond to and protect themselves against DNA damaging agents. The emerging picture from these global genomic studies is quite different from the previous concept of DNA repair, cell cycle control and induction of apoptosis as being independent processes. In fact these processes appear to form a fully integrated network. Integration of these genome-wide measure ments allows the development of specific models of response networks that could not have been detected or discerned previously. These models can be further refined and tested to improve mechanistic understanding. Specifically, the main goals of this Work shop are to enhance harmonization of protocols, to integrate databases and to increase communication between scientists in the EU and US in this field of research.
