MOLECULAR MECHANISMS CONTROLLING GENE ACTIVITY IN DROSOPHILA

The Adh region of the Drosophila melanogaster has been characterised. In particular, the physical limits of lethal(2)br22 and nocA, 2 genes which lie distal to Adh, have been defined in greater detail. The limits of lethal(2)br22 have been defined within a 12 kb genomic region by mapping the breakpoints associated with various mutations. The transcripts associated with this region is a 3.0 kb/3.1 kb doublet which is expressed throughout development with maximal levels of accumulation during the later half of embryonic and pupal development. Analysis of cDNA clones corresponding to these mRNA species indicates that they are the products of alternate splicing. DNA sequence analysis has not yet revealed any obvious homologies, but the sequence of the whole of the transcribed region is not yet complete. The expression pattern of the mRNA doublet was examined by in situ hybridization to tissue sections. The results indicate that these mRNAs appear to be expressed out of phase with the engrailed pattern, suggesting the possibility that they may be downstream genes whose expression is regulated by the engrailed product.
