Obiettivo The clinical use of engineered tissues depends upon the ability to direct cells to form specialized tissues with characteristic structural and mechanical properties. Functional implants generated in vitro would lead to the clinical use of engineered grafts tailored to (re)establish appropriate structure and function of lost or damaged native tissues. The clinical need for implants will be addressed in the specific context of the engineering of osteochondral plugs (cartilage surface at one end, bone at the o ther end, transient zone in between). The unifying hypothesis of the proposed research is that osteochondral composite grafts can be engineered in vitro by directed biophysical regulation of adult mesenchymal stem cell differentiation through biomimetic cu lture systems that provide gradients of regulatory molecules. We propose to set the differentiating stimuli through the controlled release of regulatory molecules from drug delivery systems, incorporated into a scaffold matrix. The objectives are (i) to de velop technologies for engineering stratified osteochondral tissues for reconstructions, and (ii) to improve our understanding of the mechanisms of adult cell growth and differentiation into complex tissues. The proposed application addresses a substantial clinical need of tissue-engineered reconstructions in osteochondral surfaces through the in vitro generation of functional stratified tissues, by biophysical regulation of human mesenchymal stem cells.This project fits in the Third Priority Thematic Area of Research and more precisely in the third topic of objective 3.4.1.1. Long-term interdisciplinary research into understanding phenomena, mastering processes and developing research tools. The objectives meet a socioeconomic demand in the EU and are expec ted to bring autologous and engineered tissues a step closer to clinical use.
