Dynamic Combinatorial Chemistry - selection based approach to catalysis

Obiettivo

Dynamic Combinatorial Chemistry is proving to be a powerful tool in the search for new host-guest systems. A dynamic combinatorial library consists of a mixture of compounds made by connecting building blocks through reversible covalent linkages. Addition of a template (host or guest) shifts the equilibrium in favour of a strongly binding partner at the expense of other library members. By this means a very large number of compounds are screened, and simple analytical techniques can determine suitable matc hes for the template. In the case where the template is a transition state analogue for a given reaction the host(s) produced would be expected to act as catalysts (transistion state stabilization). In this way the system itself selects the potential cata lysts and drives their synthesis. This will be exploited in the proposed research, where catalysts will be sought out through selection based methods, starting with ester hydrolysis and advancing to asymmetric catalysis and peptide cleavage.
