Novel molecular beacon technologies for the study of hiv-1 and other infectious agents

The proposed research concerns molecular beacon technology in infectious diseases in general and HIV disease in particular. This is a well-timed area with numerous possible applications and where fundamental technologies in biomedical sciences are limiting and novel technologies are increasingly in demand.

The first overall goal of this initial research project is to investigate the role of HIV-1 cellular viral load and antiretroviral drug-resistant variants on the antiretroviral treatment of HIV-1-infected individuals by using novel quantitative HIV-1 diagnostic assays. The assays have been previously constructed and evaluated in our laboratory and are based on molecular beacon and allele discriminating real-time PCR technologies that is the primary technology of our laboratory.

Our first aim involves investigating the implications of the concentration of HIV-1 DNA in peripheral blood mononuclear cell PBMC) samples ('HIV-1 cellular Viral Load') in predicting the virological response to antiretroviral therapy. In the second aim, we will quantify minor populations of HIV-1 drug resistant strains on mother-to-infant HIV-1 cohorts.

The results of the proposed studies will have a direct impact on the clinical care of HIV-1-infected individuals including Cypriot patients. The second goal is to investigate the phylogenic (subtype) distribution and spread of antiretroviral drug-resistance of HIV-1 strains from recent and long-standing patients in Cyprus.

It is hoped that the grant support of the Marie Curie International Reintegration Grant will allow the principal investigator to establish a new research laboratory in the University of Cyprus in the best possible conditions and initiate my research activities.
