The role of the Streptomyces ParB-DNA complex in chromosome segregation and regulation of DNA replication

The proposed project focuses on chromosome segregation in the mycelial microorganism Streptomyces coelicolor. Efficient chromosome segregation during S. coelicolor sporulation is dependent on partitioning protein ParB.

Using biochemical studies and subcellular localization analysis of ParB in S. coelicolor hyphae, we proved that it forms very large complexes upon binding to DNA. Here we propose further investigation of the role of the ParB complexes. We will examine an influence of the ParB-DNA complexes on chromosome structure and their role in regulation of chromosome replication.

For these studies, the origin of replication will be labelled and the replication machinery will be localised in S. coelicolor multigenomic hyphal compartments. Additionally, we aim to find an anchorage for ParB-DNA complexes followed by in-vitro and in-vivo verification of the identified interacting partners.

This study should contribute significantly to our understanding of bacterial growth and development. The proposed project is technically complementary to, and scientifically convergent with, ongoing research conducted in the host institute, which is mainly focused on Streptomyces chromosome replication.
