Structure-function relationship of TAF9 proteins: potential regulators of the survival motor neuron protein complex and targets for rational drug design

Objectif

The main objective of this proposal is the elucidation of the structure and function of TAF9 proteins and of their potential involvement in Spinal Muscular Atrophy (SMA) pathogenesis. Furthermore, the project is expected to provide useful information on th e organization of the eukaryotic nucleus. If a role of TAF9 proteins as molecular targets for SMA emerged from this project, the structure-function relationship will provide the framework for structure-assisted rational drug design for the treatment of SMA and possibly of other neurodegenerative diseases sharing common features. On an international level, the proposed research activity enhances the synergy between European centres of excellence on a research topic of wide interest, by incorporating collaborations with multiple European laboratories as an essential component. In this vein, the project aspires to initiate both future RTN and RTD Marie Curie actions and collaboration with third-country organisations like the FSMA or the MDA (both USA). On a national level, this project will represent one of the most substantial contributions of Greece to the international research effort to find a cure for SMA. The national significance of the project is underlined by the close collaboration the host institute has with academic organisations and large pharmaceutical companies, like AstraZeneca and Pfizer, thus boosting the academic and industrial competitiveness of Greece and the EU in the biotechnology sector. Another key point of this proposal is to facilitate the establishment of the researcher as an independent investigator. This will be achieved through a well-defined project that combines research methodology in the fields of biochemistry and structural biology, in which the researcher collected experience during his doctoral and postdoctoral training, and of modern molecular biology in which the researcher has specialised during his recent 30-month EU-funded mobility period
