Objective In this proposed project, single molecule Fluorescence Resonance Energy Transfer (FRET) studies on human telomerase, a ribonucleoprotein which polymerises the telomeric ends of eucaryotic chromosomes de novo, will be performed.The investigations will provide insight about telomerase structure, its dynamics and catalytic mechanism. In the first part of the project, the structure and dynamics of the RNA template of human telomerase (hTR) alone and in complex with human telomerase (hTERT) will be explored.In the second part, a methodology to investigate the telomerase mechanism by single molecule spectroscopy will be established. For the structure and dynamics studies, the RNA component of human telomerase will be labeled by hybridisation of fluorescently labeled PNA probes.
