Final Report - RENZI_FP7_IOF2007 (CryoEm structure of gamma secretase: a key component in Alzheimer neurodegenerative disease)

This is the final report of the RENZI project, which is funded under the European Commission (EC) Seventh Framework Programme (FP7). The project aimed to study the cyro- Electron Microscopy (EM) structure of gamma-secretase (g-sec) - a key component in Alzheimer neurodegenerative disease - and to provide its 3D reconstruction by using single particle EM. In the long-term, the project envisaged widening research on previous X-ray crystallography in order to study large complexes at low to medium resolution. This would enable the structure of small proteins to be determined at high-resolution and implemented by the EM techniques.

Suitable specimens for EM were prepared to further knowledge on handling electron microscopes and collecting EM images. Results suggest that location of surface cavities, their position relative to the internal chamber and membrane, offer a possible pathway for the substrate to access the active site and cleavage products to be released.
