Mid-Term Report Summary - MUSCLE-NET (Coactivator-controlled transcriptional networks regulating skeletal muscle cell plasticity)

In the first support period of our ERC Consolidator grant, the proposed milestones were fully achieved. While most of the work focused on data collection, computational analysis of some sub-projects already resulted in first publications. In particular, we discovered a novel link between co-activation of the estrogen-related receptor alpha by PGC-1alpha, which is dependent on the genomic context of enhancer and promoter regions of PGC-1alpha target genes in muscle. These insights could help in devising novel methods to selectively modulate the expression of these genes that have been found to be dysregulated in different diseases, e.g. type 2 diabetes. The next phase of the ERC grant will be marked by a shift from mainly collection to analysis of data: for that purpose, our computational expertise within and outside of our group has been significantly boosted. Finally, we are currently generating novel animal models that not only facilitate the execution of the projects within the ERC grant, but should also help other groups working on PGC-1alpha to study the regulation and function of this co-activator in vivo.
