Mid-Term Report Summary - CLOC (Cultured Liver Organoids for Investigation and Treatment of Inherited Cholestatic Diseases)

This project aims to develop novel liver regenerative models applicable to treatment development in human liver disorders. The novel liver model is being developed from organic liver scaffolds and liver cells generated from induced pluripotent stem cells (iPSc). As planned we generated iPS cells from 6 patients with genetic liver diseases and produced lentiviral vectors able to correct the gene deficiencies in the iPSc.
We have developed in house methods to generate varies types of liver cells from iPSc.
The methods to generate organic liver scaffolds have been optimised and the protocols for differentiation of the liver cells from iPSc on the scaffolds have been set up.
Novel bioreactors have been designed and bioreactor differentiation culture of the iPSc on liver scaffolds are in progress.
The novel surgical and biochemical approaches to testing cultured livers are being developed.
