Obiettivo Non-enzymatic glycosylation of proteins (glycation) is considered a main cause of nuclear cataract i.e. discoloration and increased light scatter of the lens nucleus with age. It leads to accumulation of yellow/brown pigments, appearance of non-tryptophan fluorescence and formation of cross-linked proteins. Much less attention has been given to the effects of the glycation products or derivatives on the lens membranes and their physiological properties.In addition the relation between glycation and "autooxidation" of glucose and consequently of free radicals is underscored up till now. The present proposal aims to investigate the precise chemical course of glycation and the effects on membrane intrinsic proteins and the phospholipids and cholesterol of the membranes. As such this proposal is a welcome extension of the themes studied within the Concerted action.
