Folding and stability studies of the SH3 and Gb domains

Objective



The spectrin SH3-fragment is an excellent model for investigating the energetics of minimal globular domain folding. The participants in this project will purify and study the naturally occurring variants of the SH3 domains and design some of their mutants with a view to changing fragment stability. Special attention will be paid to the mutants with variable lengths in the loops connecting the elements of secondary structure and to chimeras of the SH3.

This project also includes studies of the interaction between SH3 domains and synthetic oligopeptides modelling the structure of the functionally important recognition partners of the domain.

A multidisciplinary thermodynamic, kinetic and structural approach will be used to study folding/unfolding patterns of these species and to characterise their stability. The Russian group will purify proteins and carry out the differential scanning calorimetry and optical studies. One of the members of this group will be directly involved in mutant engineering and protein-peptide modelling studies by joining the German group.
