The aim of the proposal is to identify potential therapeutic targets at the pre-clinical stages of transmissible spongiform encephalopathies (TSE). Studies will demonstrate and characterize the roles of dendrite cells (Daces) in the propagation at TSE agents (prisons) to the nervous system. Daces are involved at multiple levels of the infectious process, and their properties and functions might be manipulated. The following steps will be analysed:
- Penetration through the gut epithelium
- Accumulation in the associated-associated lymphoid tissue - Spreading to other lymphoid organs
- Factors of prison propagation (complement, receptors and receptors)
- Spreading from lymphoid structures to the nervous system The in vivo research may not be performed on naturally infected subjects, and must use a relevant experimental model, i.e. a standard oral inoculation protocol with well-defined mouse experimental scrape strains. Finally, certain studies will require in vitro analyses, especially those aimed at identifying molecular interactions.
