High frequency stimulation of the subthalamic nucleus: a therapeutic approach to parkinson' s disease

Parkinson's disease (PD) is a neurodegenerative illness with high prevalence and long course characterized by severe motor and cognitive impairments. Clinical trials have recently shown that chronic high frequency stimulation (HFS) of sub thalamic nucleus (STN) alleviates Parkinson Ian motor symptoms and elodea-induced complications at advanced stages of PD. However, little is known about the functional mechanisms of this new therapeutically tool, its long-term motor effects and consequences on cognitive functions. The aim of this project is thus to address these issues. Behavioural consequences of STN HFS will be specified in terms of motor and cognitive functions in PD patients and in animal models. Biochemical and cellular events underlying the behavioural effects of STN HFS and neuropath logical factors that may govern its efficacy will be investigated in animal models. The results are expected to have major socio-economic impacts by improving indication of STN HFS in the course of PD.
