Biotechnological process control of chlorinated solvent and air emission

The objectives of the study are to assess the feasibility of using bioreactors for treatment of volatile hylogenated aliphatic compound(VHAL) contaminated water and air.

The focus of the study will emphasize degradation of trichloroethylene(TCE) by strain G4, but the ability to biodegrade other chlorinated aliphatic compounds will also be investigated as well as screening for other microorganisms capable of degrading VHALs. Laboratory scale bioreactors will be designed and constructed for treatment of water and air contaminated with TCE. Kinetics of biodegradation of TCE by strain G4 will be assessed in both of these systems. Phenol will be tested for its efficacy in maintenance of TCE degradation and kinetics of removal under simulated field conditions.

Batch studies will be performed with strain G4 to determine which other target VHALs might be degradable. Specific VHALs that will be assessed for biological detoxification will include tetrachloroethene (perchloroethylene(PCE)), trichloroethene (trichloroethylene(TCE)), 1,1 dichloroethene (vinylidene chloride), and cis and trans-1,2-dichloroethene 1,1,1-trichloroethane (TCA). Recent results indicate excellent potential for biological detoxification of VHALs.

Specific objectives of the study include :
development of a bench scale bioreactor for the treatment of TCE contaminated water using sterile, defined conditions (G4);
assessment of its utility by treatment of nonsterile TCE contaminated groundwater;
development of a bench scale bioreactor for the treatment of TCE contaminated air using parallel flow reactor design;
confirmation that TCE removal in both reactors is biological and results in mineralization, by sodium azide poisoning and carbon-14 mineralization experiments, respectively;
determination of the ability of strain G4 to degrade other target VHALs;
and to screen environmental samples for unique microbial degradation systems.
