Identification and quantitation of species in marine products.

Cel

The aims of the projects are:
to standardize as far as possible the techniques of analysis in the respective laboratories;
to investigate the use of densitometry, image analysis and computerization of data as developments of the basic separation and staining techniques;
to solve, if possible, the problem of identifying certain closely related species;
and to develop methods for the quantification of relative amounts of different species in multispecies products.
The principle of identifying species of raw fish by electrophoresis or isoelectric focusing (IEF) on the water soluble protein is well established. There can be problems in interpretation of closely related species as the profiles vary to some extent. The reasons for this variability are not well understood but they could be influenced by biological condition and fishing ground. Further information is needed on the degree of variability of the profiles of fish species of commercial importance. At present, no method exists which precisely quantifies the amount of a known fish species in a product. The project is being set up to standardise as far as possible the techniques of analysis in the participating laboratories, to solve the problem of identifying certain closely related species, and to develop methods for the quantification of relative amounts of different species in multispecies products.

An initial comparative study of the analytic systems used by the participating laboratories was carried out to determine which parameters of IEF had to be standardised for fish species identification and which steps in the analytic procedures were not so critical for the reliability of the results.
