Drug Design for Cardiovascular Diseases: Integration of in Silico and in Vitro Analysis

Recent technological advances have greatly increased the capability to investigate complex molecular interactions that occur in the onset of disease in order to identify genetic alterations and to screen new drugs.The first critical step of the drug deve lopment process is the identification and the validation of drug targets for the therapeutic application. At present about 500 molecular targets have been identified and validated.This is the basis of putative drug molecules that are able to interact wit h such targets.Recent advances have greatly speeded up the screening of the new drugs, however the number of resulting molecules which are not pharmacologically active is still extremely large. The optimisation of screening depends on the integration of in silico and in vitro analyses. The screening of targets is extreme difficult for multifactorial diseases, which are characterized by complex synergies between genetic and environmental factors.Cardiovascular diseases (CAD) are one of the major group of multifactorial diseases and are one the main causes of mortality and morbidity in most developed countries. The aim of this project is to discover new factors involved in atherogenesis and their related CAD and set up systems to select and test molecules that are pharmacologically active for the treatment of CAD.The project combines the computational (bioinformatics,cheminformatics and bioengineering) and the experimental (clinical medicine and biology) expertise.Clinical information drives bioinformati cs analysis.The results are the basic knowledge for cheminformatic analysis. Subsequently putative targets will be synthetised by industrial partner and tested on specific experimental models. These information,combined with physiological parameters, are used for the development of an in silico disease. A GRID infrastructure will be used to screen potential drug candidates. In addition,the project will provide a specialized portal for CAD.
