Course on high-throughput microRNA profiling, Heidelberg, Germany
It will be composed of 60% hands-on laboratory work and tutorials and 40% lectures. The lab sessions will cover topics such as quality assessment and quantification of total RNA, setup of the complete microarray-microRNA profiling experiment, synthesis of complementary DNA followed by microRNA specific amplification, data analysis by different algorithms, normalisation approaches and troubleshooting.
Lectures, on the other hand, will discuss:
- approaches to microRNA profiling;
- sample preparation;
- experimental design/setup;
- quality controls for total RNA;
- equipment/instrumentation options;
- design of quantitative real time polymerase chain reaction (qPCR) experiments;
- design microarray microRNA experiment;
- data analysis, normalisation and interpretation;
- correlation of transcriptome profiling with microRNA profiling data;
- correlation between various microRNA profiling approaches;
