Objective
In vivo, cross-presentation of exogenous antigens has mostly been associated with dendritic cells. This process, which does not depend on the degradation of newly synthesized proteins, allows for the acquisition and presentation of peptides derived from exogenous antigens on MHC class I molecules. Better understanding of the mechanism of cross-presentation is essential, first because it appears crucial for priming CD8-positive T cell responses, and second because it has implications for various issues such as transplantation, viral infection, vaccination, autoimmune diseases and cancer.
Recent reports have highlighted crucial aspects of the cellular mechanisms involved in cross-presentation. However, our understanding of this phenomenon remains incomplete. Here, we propose to study cross-presentation of exogenous antigens by confocal video microscopy and biochemical approaches, with a focus on the cross-presentation of tumour specific antigens of the MAGE-type.
This study will be realized through a collaboration between two partners: the laboratory of Pr Cresswell in the Section of Immunobiology in Yale University Medical School, and the Cellular Genetics Unit of the Christian de Duve Institute of Cellular Pathology.
