Objective The compound microscope is one of the most widely used tools in medicine and it has numerous other uses in analytical science. In the last decade attention has returned to the attributes of wide field microscopy using coherent illumination and holographic recording techniques largely due to the increased capabilities of digital cameras and computing hardware. The main advantages of holographic microscopy are: a greater depth of field (few hundred microns at high resolution); the aberration and distortion in troduced by poor quality windows can be compensated; and the ability different output type images by software manipulation. The overall aim of this work is to demonstrate coherent imaging and apply new methods to analyse the data recorded by a coherent mic roscope. It includes the conversion of an existing inverted microscope to demonstrate the capability of coherent imaging with a view to bio-medical applications. This project also proposes to investigate flexible analysis methods to post process coherent i mages. We will implement methods to provide the output data as Differential Interference Contrast and Hoffman images and investigate software that will increase the depth of field indefinitely (at the expense of resolution). The final objective of this wor k is to investigate methods to calculate the three-dimensional distribution of refractive index for the purposes of cell visualisation and classification.
