Molecular control of brain regionalization and neural cell diferentaition by planar inductions

Objective

This proposal deals with the role of Fgf8 in regulating regionalisation of the vertebrate brain during embryogenesis and its potential role as a regulator of function in the mature brain. In embryos Fgf8 is a primary signal involved in brain regionalisation and pattern formation. Localised Fgf8 expression in co-operation with Wnt, Bmp and Shh signals promotes localised cell fate determination and long range patterning of surrounding areas. We propose to study the role of FGF8 dependent Ca2+ signalling in this long range integrated signalling phenomenon. The localised sites of Fgf8 expression persist at least until birth and correspond to brain nuclei that regulate brain state including the Supra-chiasmatic Nucleus and Bed Nucleus of the Stria Terminalis that function as circadian pacemakers to determine daily activity patterns. We propose to study what role Fgf8 might play in regulating these clock regions and to what extent adult function represents a continuation of embryonic function.
