Phospholipid and glycolipid recognition, interactions and structures by magnetic resonance

Objective

In this specific targeted research project we intend to use cutting-edge nuclear magnetic resonance (NMR) techniques to develop a sensitive, rapid and integrated approach to understanding the mechanisms of lipid binding proteins. The project will generalize and accelerate the efforts to characterise a range of peripheral membrane proteins, thus providing new insight into an important class of proteins which play a key role in regulating complex cellular pathways.

Model membranes will be designed to mimic various types of cellular membranes for selected protein modules which will be prepared by specific advanced tailored protein expression methods. The generation of focussed lipid molecule libraries will allow definition of membrane targeting specificity, and the synthesis of novel spin labels and micelle-forming compounds will facilitate the measurement of the angles and depths of bilayer insertion.
