Novel strategies for the total synthesis of bioactive macrocyclic natural products


The synthesis of natural compounds with biological activity is one of the most important areas in organic chemistry, and also for the agrochemical and pharmaceutical industry.

Polyketides are the most abundant medicinal agents among natural products. Structural diversity and wide scope of bioactivities are typical of the group. This proposal is focused upon the synthesis of resorcylate bio-active natural products and the underlying synthetic methodology.

The objective of this research project is the total synthesis of two resorcylate natural products with potential bio-activity, the important anabolic, estrogenic and antibacterial macrolide zearalenone and the potent antifungal agent 15G256b (previously not synthesized), through concise and simplified bio mimetic-like routes.

The methodology that will be used for the development of the synthetic route will be based on lactone macrocyclisation using enyne metathesis reaction and calcium (II) catalytic transesterification as key steps, also studying the scope o f these reactions in the synthesis of polyfunctionalised targets.

The development of the project will contribute positively to my career development as a scientist, offering me the key experiences to mature and enhance my career in the future as an independent researcher.

It will be beneficial for the objectives of the European Research Area because it will qualify me to develop my future career inside the European Community completing my scientist training with trans-national mobility, a strategic goal for the European Policy training objectives.
