The technologies of biological membranes

Objective

Many will argue that this will be the decade which unlocks the power of the biological membrane. It is the key element in the organisation and regulation of cells and therefore is of fundamental importance to biology. Allied to this, the ability to influence membrane behaviour (eg. by therapeutic intervention) or to harness the principles and components of membranes to technological applications (eg. biosensors) will usher in a new era of exploitation and innovative commercial development. This application is submitted in order to introduce into a rapidly emergent University in Ireland, a range of projects and their accompanying technologies associated with biological membrane science. It will at the same time bring to the whole of Ireland a major research unit dedicated to the study of biomembranes, a field which is under-represented in the Irish research portfolio. In so doing, it will inform and stimulate the community, provide a new focus for research and, most importantly, create a new skills base. This latter aspect is particularly relevant to the development of the private health sector where the greatest proportion of research invested is currently directed towards the membrane.
The protein systems to be studied are involved in cellular regulation (eg. G-protein coupled receptors, ion channels, lipocalin receptors) and membrane protein interactions and structure, many associated with defined diseases or human dysfunction. The areas of research will be structure/function relationships, protein interactions, protein folding and assembly, structural characterisation and cell biology.
The technologies to be employed include membrane protein expression, purification and reconstitution, membrane-directed 2-hybrid syste4ms, surface plasmon resonance, microphysiometry and the proteomics of membrane systems.
