Molecular Sensors and Dosimeters for the Optical Detection of Ions and Molecules

Objective

The present research project is aimed to the design of molecular devices suitable for the optical detection of electrically charged and neutral guests. In particular, we intend to prepare dual responsive sensing devices, which are able to communicate to the outside, the occurrence of the recognition event through two distinct and independent channels:
(i) colour change and
(ii) modification of the fluorescence emission. This goal will be achieved by using as a signalling subunit a molecular fragment, which are both a chromospheres and a fluorophore. This dual responsive feature will be especially useful for the real space and the real time determination of analyses in systems like the whole cell and tissues, when investigated by optical co focal microscopy. Sensing devices will be designed for the following classes of analyses: metal ions, anions, amino acids, and small molecules like NO and CO. Two distinct paradigms will be followed for the design of the molecular sensor:
(i) the covalent linking approach (the receptor moiety and the signalling subunit are covalently linked together, either directly or through a spacer);
(ii) the chemo sensing ensemble approach, which involves the weak and reversible interaction of the receptor with an optical indicator, to be displaced by the desired analyse. Either approach will be used depending upon the nature of the substrate. For the detection of transition metal ions, multidentate legends will be used as receptors. The nature of the chelating subunit, whether open or cyclic, will define the type of sensing device: with an open-chain receptor, metal binding will occur in a fully reversible way, thus giving rise to a sensor, if the receptor is cyclic, the metal will be included irreversibly into the macro cyclic cavity, thus producing a dosimeter, i.e. a permanent device, which accumulates the dose and sums up the signal.
