Elucidation of light signaling pathways that control the photoperiodic induction of flowering

Objective

Plants often flower in response to environmental signals. Day length controls flowering in many specie,s enabling seasonal control of flowering and adaptation to life at particular latitudes. In the model species Arabidopsis, a regulatory pathway required for the initiation of flowering has been identified and many of the components are conserved in distantly related species, suggesting its importance throughout the plant kingdom.

In Arabidopsis the CONSTANS (CO) and FT proteins play an important role in this pathway, and strongly promote flowering. CO activates the transcription of FT, but only under long day lengths. This occurs because CO protein is activated by exposure of plants to light, and the time of CO transcription is controlled, so that the protein is present in light only under long days.

The trainee proposes to define the molecular mechanism by which CO protein is activated by light, by identifying mutations that prevent this and then cloning the affected genes. This will be facilitated by materials available in the host laboratory: plants carrying a fusion of the FT promoter to luciferase and CO expressed from a viral promoter.

Using these materials, mutations will be identified that reduce activation of FT by CO by identifying plants with reduced luciferase expression. The genes will be isolated by map-based cloning, and the function of the proteins in light signalling investigated. The host laboratory has the infrastructure required for these studies, including high-throughput luciferase detection systems.

Training of the applicant will occur within an international institute and a host laboratory experienced in training foreign postdoctoral fellows, and will include laboratory-based work, oral presentations and seminar programmes.
