Objective
The malignant tumours comprise a mixture of transformed cells, normal cells and extracellular matrix. The normal cells within a tumour, predominantly endothelial cells, fibroblasts, lymphocytes and macro-phages, contribute to tumour growth and progression. Tumour associated macrophages, TAM, are especially important in this regard. The aim of this project is to study interactions between TAM and tumour cells that may enhance tumour cell invasiveness, and other processes in tumours, such as angiogenesis. The researcher has obtained novel data suggesting that soluble signals that pass between human macrophages and tumour cells can enhance the invasive capacity of tumour cells in a process that involves matrix metalloproteases.
The researcher obtained a competitive fellowship to visit the host laboratory from October 2002-January 2003. Using techniques and expertise of the host laboratory he demonstrated a role for the inflammatory cytokine tumour necrosis factor-alpha (TNF-a) in the interactions between TAM and tumour cells. The aim of the proposed project is two fold. First the researcher wishes to investigate the in vivo relevance of his observations using mouse cancer models and transgenic mice that are established in the host laboratory.
Second, the researcher wishes to use state-of-the-art expression array facilities in the host Institute, in collaboration with proteomics facilities available in his home laboratory, to further investigate the mechanisms by which macrophages enhance tumour cell invasiveness .It is hoped that the data obtained from this fellowship application may lead to novel therapeutic and diagnostic/prognostic strategies in patients with invasive cancer, and establish a continuing collaboration between the host laboratory and Gottingen.
