Repressive histone methylation networks

Objective

Epigenetic regulation of higher-order chromatin structure is currently an area of intense research. Chromatin structure affects fundamental biological processes e.g. regulation of gene expression during development, genome stability, various diseases and cancer. Key regulators of these processes are histone methyl transferases.

One of the objectives of the proposed research project is to identify novel regulators and components of heterochromatin in higher eukaryotes. We plan to map mutations in Drosophila that are involved in epigenetic gene silencing using rapid SNP analysis. Mammalian homologues will be identified and characterised in mouse cells. Localisation of EGFP-fusions will be studied. Employing immunostaining and RNAi knockdown, the importance of these genes in the maintenance of heterochromatin will be analysed.

Another objective is to explore protein-protein interactions among components and regulators of heterochromatin. Mammalian heterochromatin components will be fused to affinity tag s and protein complexes harbouring them will be isolated from cell cultures. Binding partners will be identified by mass-spectrometry.
