Brain endothelial cell-based gene therapy of neurodegenerative diseases

Objective



Neurodegenerative diseases, like Alzheimer's, Parkinson's or Huntington's diseases, constitute a major health problem in Europe. There are no therapies for these diseases except in the case of Parkinson's disease where current pharmcological treatment can limit for a time the evolution of the symptoms. However, some neurotrophic factors have been identified as drug candidates. Neurotech's core technology is based on the use of brain endothelial cell lines as cellular vectors for drug delivery to the CNS. Accordingly, the present programme is focused on the production of gentically engineered brain endothelial cell lines secreting neurotrophic factors under the control of viral or housekeeping gene promotors identified by NeuroSearch. The therapeutic efficacy of these cell lines will be evaluated by cerebral transplantation in rats with experimentally induced neurodegenerative diseases. Among the criteria assessed will be the survival of susceptible neuronal populations and the improvement in animal behavior at various times after transplantation. As a therapeutic approach of neural transplantation in Parkinson's disease, the feasibility of grafting human fetal neurons is now clinically well established. However, this strategy, dependant on a constant supply of human fetal tissues, raises important ehtical and practical issues. Alternatively, the present programme, based on the use of stable cerebral endotherlial cell lines, could contribute to the further development of a novel therapeutic strategy for neurogenerative diseases.
