Objective
Aging of the popoulation by increasing life expectancy is making the problem of age-related dementia increasingly important. Senile dementia is becoming a serious problem not only for family members including the patient but also for modern society. the existing drugs have been effective only for the complicated symptoms of related diseases. Hence, nex drugs effective for the essential pathology are required. Since 1990 the prime proposer has developed different types of highly effective compounds inhibiting proly endopeptidase (PEP), on of the putative key enzymes inducing the senile plagues formation. Two initial patent have been filed. The main goal of the research proposal is to tailor our already in vitro powerful inhibitors to be useful diagnostic and therapeutic compounds in vivo and clear the way for development of marketable drugs out of this invention. The following main research tasks will be approached on the molecular, cellular, physiological and behavioral levels: - elucidation of the molecular structure of brain PEP and their catalytic properties - investigation of the neurofunctional consequences of an inhibition/lack of PEP (application of highly effective inhibitors, antibodies and antisense oligonucleotides) - study of the physiological effects of sequence modified of neuroactive peptides and PEP inhibitors on long-term potentiation in vitro and in vivo - developing of organ and tissue-senstive inhibitors of PEP - pharmacokinetic and pharmacodynamic studies on PEP inhibitors To investigate the functional role of proline-specific peptidases on molecular, cellular and physiological level needs the combination of European skills and knowledge in neurobiology. Since about one third of the population in the age of 65 is suffering onset of Alzheimer's disease, the market potential of a drug developed on the specific mechanism ofy-secretase inhibition is to be estimated very high. The interest of our SME is to sell license of the newly developed compounds to big pharmaceutical enterprise or to pursue own clinical phase studies together with SME in Europe.
