Gene-Regulatory DNA-binding Drugs

Objective



The proposed project concerns a new approach to drug therapy, gene regulatory DNA-binding drugs. Such agents are conceived to modify disease processes by altering the expression of the genes associated with them. The envisaged molecular mechanism of action involves the displacement of an appropriate transcription factor from the promoter region of the target gene by a low molecular weight(<1000Da) sequence-sepcific DNA-binding compound. Potential applications exist in the treatment of viral infections, cancer, and various other diseases. The proposed research will result in the development of a library of sequence-specific DNA-binding molecular fragment, which will comprise the building blocks from which the gene-regulatory drugs will subsequently be constructed. Structural studies of DNA and DNA:Ligand interactions usig NMR spectroscopy will provide a starting point for rational molecular design and combinatorial chemistry. Designed compounds will be evaluated in a proprietory DNA-binding assay system, by DNA footprinting, and in various in vitro functional assays.
