Mimotopes of carbohydrate antigens as possible vaccines against bacterial and parasitic infections

Objective



We have recently demonstrated that peptide mimics, i.e. mimotopes, selected through the screening of phage-displayed peptide libraries using a protective monoclonal antibody directed against a carbohydrate antigen of an intestinal pathogen, induce upon immunization anti-carbohydrate antibodies. Therefore, mimicking of non-protein ligands can be achieved through screening random peptide libraries where the specific sequences are identified using a purely combinatorial approach, without any structural information of the original target. These data suggest that a novel class of subunit vaccines to polysaccharide antigens, based on the use of selected mimotopes as immunogens, might be developed. The proposed project plans to evaluate this mimotope-based subunit vaccine strategy for two human intestinal pathogens. Its unique and essential feature is to propose the development of anti-polysaccharide vaccines which do not require knowledge on, purification or synthesis of the original target. The proposed project cannot be carried out by a single research group. The best chance of success is through the development of a transdisciplinary network that involves groups with expertise in molecular genetics of and immunity to pathogens, in phage-displayed peptide technology, in delivering of immunogens for the induction of systemic as well as mucosal immunity, and in adjuvant molecules. The transeuropean network we propose here is more than the simple sum of its composing groups of experts. It proposes a synergistic and creative collaboration which represents the only way to efficiently tackle modern vaccinology.
