Integration at pilot scale of high cell density bioreactor operation with ultra fast on-line downstream processing

Objective



This project aims at validating - at pilot scale - a continuous process whereby heterologous glycoproteins secreted from mammalian cells propagated in continuously stirred tank reactor are extensively purified by an uninterrupted downstream processing.

The original aspects of the Demonstration are:
- integration of high cell density cultivation with ultra fast on-line downstream processing - validation of sonoperfusion at 8L- and 20L culture volume scale - validation of the "high cell concentration per unit culture volume" concept - use of state-of-the art support for Expanded Bed specific Adsorption (EBSA) - use of specific solid phase screening methods for the selection of chromatographic ligands.
