Development of hyphenated analytical chemical techniques and application to the study of reactive drug metabolites and normal and abnormal plasma lipoproteins

Objective

The major objectives of this research project are to develop new and improve hyphenated techniques (e.g. LC-NMR-MS, CE-NMR-MS, CE-MS-MS) for the use in
- investigation of reactive drug metabolites (drug allergy)
- investigation of plasma proteins (diagnostic tool)

The overall aim of the proposed RTD project is to further develop and apply hyphenated methods involving separation techniques [high-performance liquid chromatography (HPLC) and capillary electrophoresis (CE)] and nuclear magnetic resonance (NMR) and mass spectrometry (MS) for the study of i) reactive drug metabolites (in particular acyl glucuronides) in relation to their impact on drug allergy and other toxicological consequences and of ii) plasma lipoproteins in order to develop better instrumental techniques for use as diagnostic tools in relation to cardiovascular diseases and cancer. An important aim is also to establish a network for the training of researchers in the analytical chemical and adjacent techniques developed in this RTD project. The proposal gives the background for the two research areas in which we think our research will provide new knowledge. The new knowledge on hyphenation will be exploited by Bruker Analytische Messtechnik and will ultimately be published in international journals. The proposal describes how the objectives will be fulfilled by completing a number of task :
1. Development of methods in HPLC-NMR (including chiral separations), micropore HPLC-NMR and CE-NMR
2. Development of HPLC-NMR and CE-NMR methods for lipoprotein analysis.
3. Analysis of lipoprotein and oxidised lipoprotein composition in normal and abnormal plasma by NMR.
4. Synthesis of model and drug glucuronides.
5. Use of 1H and l9F NMR and HPLC-NMR to derive transacylation kinetics.
6. Quantitative structure-activity studies on transacylation using computional chemistry properties.
7. Characterisation of drug glucuronide/protein condensation products.
