Enzymatic synthesis of chiral drugs: cyclopirrolones and 1,4-dihydropyridines

Objective



The scientific and technical objectives of this research proposal concern pharmaceutical registration of chiral drugs: (+)-zopiclone and 1,4-dihydropyridines. Although it's very well-known the influence of the drug stereochemistry in their activity, up to 1994 most of the chiral drugs are marketed as a racemic mixtures. The biological activity, bioavailability and metabolism of a drug can be very different for each one of the enantiomers of a chiral drug. The growth of the demand of chiral drugs has stimulated the research of new roads for the synthesis of pure enantiomers with therapeutical activity. We are still at the begining of a long way that may lead to the replacement of racemic drugs by the more selective and/or potent and/or safe single enantiomer. Research is required to synthetise the chiral drugs, studie the activity, bioavailability and selectivity of optically pure enantiomers . Also required the optimisation of experimental conditions of scaie-up process.
