Determining European standards for the safe and effective use of phytomedicines

Objective



The objective is to study the effect of purified lectin substances as well as the toxophoric A-chain and the sugar-binding B-chain of the hololectins on the cytotoxic activity of human leukocytes (effector cells) against cultured human lung carcinoma cells (target cells). In the framework of the proposed study the following experiments in-vitro are planned:
i direct effect of the purified mistletoe lectins and subunits on the target cells,
ii on the natural cell-mediated cytotoxicity of the effector cells, iii combined effects of the ionizing radiation or lipopolysaccharides and mistletoe lectins on the natural cell-mediated cytotoxicity of the effector cells,
iv cytokine production by and release from the effector cells upon the treatment with the purified mistletoe lectins as detected in ELISA and
v expression of cytokine receptors (for instance TNF- receptors) on the surface of monocytes, macrophages upon the treatment with the purified mistletoe lectins.
Moreover cytogenetical studies should be carried out:
i of selected lectins on human lymphocytes,
ii develompent of tests for individual sensitivity against the lectin preparations,
predictive assays on the possible reactions after lectin treatment iii simultaneaous studies on cytological and cytoaenetic assays, iv studies on the antioxidant capacity of preparations through treatment of blood samples with ionizing radiation in the presence of lectin preparations.
