Objective
PLASMIDS OF THE PKT GROUP TRANSFERRED IN Z.MOBILIS BY CONJUGATION WITH THE HELP OF CONJUGATIVE PLASMID LIKE RP4. TRANSFER FREQUENCIES WERE IN THE RANGE OF 10 TO THE POWER OF MINUS 5 - 10 TO THE POWER OF MINUS 6 CONJUGANTS PER RECIPIENT CELLS. PLASMIDS PKT210, PKT248, PKT212 AND PGSS33 WERE STABLY MAINTAINED IN Z MOBILIS. PLASMIDS FROM Z.MOBILIS SHOWED THE SAME MOBILITY ON AGAROSE GEL ELECTROPHORESIS THAN THE ORIGINAL ONES FROM E.COLI AS WELL AS RESTRICTION ANALYSIS. THE USE OF THESE VECTORS AS CLONING VEHICULES IN Z.MOBILIS IS SUGGESTED. ETHANOL PRODUCTION, IN CONTINUOUS CULTURE, FROM SUCROSE WAS STUDIED AND OPTIMIZED. HIGH ETHANOL CONCENTRATION (AROUND 60 G/L) AND PRODUCTIVITIES (40 G/L.H) WERE OBTAINED USING A FLOCCULENT STRAIN IN A FERMENTOR WITH AN INTERNAL SETTLER. HOWEVER, THE ETHANOL YIELD WAS ONLY 80 % BECAUSE OF LEVAN FORMATION. THE CYTOPLASMIC AND OUTER MEMBRANES OF Z.MOBILIS WERE SEPARATED BY CENTRIFUGATION ON A DISCONTINUOUS SORBITOL GRADIENT. THE PROTEINS PATTERN OF THE TWO MEMBRANES HAS BEEN DETERMINED.
