THE ROLE OF DIFFERENT PATWAYS OF METABOLIC CONVERSION IN THE ASSESSMENT OF THE MUTAGENIC AND CARCINOGENIC POTENTIAL OF ENVIRONMENTAL MUTAGENS

Objective

ELABORATION AND COMPARATIVE EVALUATION OF TEST SYSTEMS FOR THE ASSESSMENT OF GENETIC EFFECTS OF ENVIRONMENTAL CHEMICALS AND OF THE PRODUCTS OF THEIR METABOLIC TRANSFORMATION.

THE SCOPE OF THE PROJECT IS THE INVESTIGATION OF IN VITRO ANS IN VIVO SYSTEMS PERMITTING FURTHER ELUCIDATION OF THE ROLE OF METABOLISM WITH REGARD TO THE MUTAGENIC AND CARCINOGENIC ACTIVITY OF ENVIRONMENTAL CHEMICALS.

IN PARTICULAR, THE INFLUENCE OF DETOXIFYING ENZYMES, SUCH AS EXPOXIDE HYDRATASE AND GLUTATHION-S-TRANSFERASE, AND OF INHIBITORS OF SUCH ENZYMES, WILL BE STUDIED IN VITRO USING SCHISTOSACCHAROMYCES POMBE AS TEST SYSTEM. THESE EXPERIMENTS WILL BE COMPLEMENTED BY APPROPRIATE IN VIVO STUDIES.

TEST COMPOUNDS WILL BE LOW MOLECULAR COMPOUNDS WITH DIFFERENT CHEMICAL STRUCTURES, IN PARTICULAR THE REFERENCE MUTAGENS SELECTED FOR THE COORDINATED EEC-PROGRAMME.

FURTHERMORE, THE PHENOMENA LINKED TO THE IN VIVO FORMATION OF NITROSAMINES AND THEIR DIRECT OF INDIRECT MUTAGENIC AND CARCINOGENIC ACTIVITY WILL BE STUDIED. FOR THIS RESEARCH, THE HOST MEDIATED ASSAY USING S. POMBE (INTRAVENOUS OR INTRAGASTRIC APPLICATION) WILL BE USED, IN COMBINATION WITH THE EXAMINATION OF GENOTOXIC EFFECTS IN VIVO (E.G. CHROMOSOMAL ABERRATION).
