MECHANISMS OF CADMIUM TOXICITY

Objectif

TO INVESTIGATE THE EFFECTS OF CADMIUM ON SOME SPECIFIC FUNCTION OF ZINC, COPPER AND IRON AND TO ATTEMPT TO ELUCIDATE THE MECHANISMS WHICH THESE EFFECTS OCCUR.

THE MOLECULAR MECHANISMS UNDERLYING THE TOXIC EFFECTS OF CADMIUM ARE LARGELY UNKNOWN AND ARE INVESTIGATED IN THIS PROJECT, AS FOLLOWS:

1) THE PRODUCTION OF AN ANTISERUM TO CADMIUM-THIONEIN AND DEVELOPMENT OF A RADIOIMMUNOASSAY FOR THE PROTEIN COMPLEX;

2) AN INVESTIGATION OF THE POSSIBLE DISTURBANCES BY CADMIUM OF THE VITAL METABOLIC FUNCTIONS OF ESSENTIAL METALS.

THE FOLLOWING STUDIES IN EXPERIMENTAL ANIMALS ARE UNDERTAKEN:

1) PRODUCTION OF ANTISERA AND DEVELOPMENT OF A RADIOIMMUNOASSAY FOR METALLOTHIONEIN;

2) STUDY OF THE EFFECTS OF CADMIUM ON MEMBRANE LIPID PEROXIDATION, MICROSOMAL FATTY ACYL-COA DESATURASE SYSTEMS, MICROSDOMAL CYTOCHROMES P-450, P-455 AND B5 MIXED FUNCTION OXIDASE ACTIVITIES;

3) DETERMINATION OF BLOOD LEVEL CHANGES OF METALLOTHIONEIN AND CADMIUM, UP TO THE DEVELOPMENT OF KIDNEY DAMAGE;

4) AN INVESTIGATION INTO THE POSSIBILITY THAT DIETARY SUPPLEMENTATION WITH ZINC AND COPPER MAY PREVENENT THE ONSET OF SOME TOXIC EFFECTS.
